表觀遺傳學(xué)是指基于非基因序列改變所致基因表達(dá)水平變化。表觀遺傳的現(xiàn)象目前已知的包括DNA甲基化、基因組印記、母體效應(yīng)、基因沉默、核仁顯性、休眠轉(zhuǎn)座子激活和RNA編輯等。
其中組蛋白(Histone)的修飾作用是表觀遺傳學(xué)研究的一個(gè)重要領(lǐng)域。Histone H3與H2A、H2B和H4一起參與真核細(xì)胞中染色質(zhì)的結(jié)構(gòu)。組蛋白H3通過(guò)承受幾種不同類(lèi)型的表觀遺傳修飾,參與并影響許多細(xì)胞進(jìn)程,如轉(zhuǎn)錄激活/失活、染色體包裝、DNA損傷/修復(fù)。這些修飾,包括甲基化、瓜氨化、乙?;⒘姿峄?、小泛素相關(guān)修飾化、泛素化及二磷酸腺苷核糖基化。主要發(fā)生在組蛋白H3的N末端尾區(qū)域上,這導(dǎo)致核小體結(jié)構(gòu)重構(gòu)成更容易接近的開(kāi)放構(gòu)象的轉(zhuǎn)錄復(fù)合物。在大多數(shù)物種中,組蛋白H3主要在賴氨酸9,14,18,23和56處乙?;? 在賴氨酸4,9,27,36和79處甲基化; 并分別在ser10,ser28,Thr3和Thr11磷酸化。組蛋白修飾的深入研究能更好地了解細(xì)胞過(guò)程的表觀遺傳調(diào)控。
